A total of 16 teams will compete in the towns throughout this edition, starting in León and finishing in Ponferrada. This was explained by the Director General of Sports, María Perrino, during the presentation of the competition, which is now in its 35th year.

Coinciding with the 2021 celebration of the Jacobean Year, the 35th edition of this sporting event, organized by the Cadalsa cycling club and sponsored by the Regional Government of Castile and León, will cover unique sections of the Camino de Santiago as it passes through León. The cycling event will consist of a challenging stage that will cross the province of León.

The Director of Sports recalled that the 35th edition of the International Cycling Tour of Castile and León was unfortunately disrupted as a result of the COVID-19 pandemic, so it was suspended approximately one month before its celebration. This year's route will be linked to the Camino de Santiago, an icon of Spanish and Castile-León cultural heritage, and will traverse sites and gems of the Camino de Santiago as it passes through Castile-León. The route starts in the city of León, passing through sites that showcase the region's natural and cultural heritage. The finish in Ponferrada will reveal the winner of the 35th Vuelta Ciclista a Castilla y León.

A total of 16 teams will participate, with up to four UCI World Teams.
